DIAGAST developed a new test DC-LYS to perform Direct Antiglobulin Test, completely adapted to QWALYS 3 System. DC-Lys is an in vitro diagnostic medical device (IVDMD) for professional use to perform the direct antiglobulin test.
DC-Lys should be only used in combinaison with MagLys, DiluentLys, ScreenDiluent 2, NanoLys and Mag-Plates uniquely in EM® Technology on fully automated machine QWALYS®
.
This test shows the in vivo sensitization of the human red blood cells by some IgG anti-erythrocytes antibodies and / or by component C3d of the complement system.
The monoclonal anti-IgG antiglobulin present in the microplate wells of DC-Lys reveals the presence of antierythrocyte IgG type antibody fixed on the red blood cells. The monoclonal anti-C3d antiglobulin présent in the well of the DC-Lys microplate shows the presence of the component C3d of the complement system fixed on the red blood cells. This revealing is translated by a positive reaction with the birth of a more or less important veil of red blood cells in the bottom of the well.

Human red blood cells have been sensitized in vitro by anti-erythrocytic antibodies of IgG type and/or by the C3d molecule of the complement. They have been tested on DC-Lys in EM® Technology on QWALYS®. Results have shown a specific recognition at 100% of sensitized red blood cells tested. These performances have been validated on 3 different batches of DC-Lys and a reproducibility inter and intra-essays has been found.

The direct antiglobulin test is used to detect the presence of immunoglobulin and/or complement on the surface of red blood cells. This test is helpful in the work-up of autoimmune hemolytic anemia, drug induced hemolytic anemia, hemolytic disease of the newborn, and hemolytic transfusion reactions.
